ÖZET
Bu çalışmada, siklofosfamid (SK) prenatal maruziyetin sıçan beyincik üzerindeki olumsuz etkilerinin araştırılması amaçlandı. Ayrıca ginkgo biloba'nın (GB) serebellar dokularda SK'nın neden olduğu olası biyolojik değişikliklere karşı koruyucu etkileri de değerlendirildi. Bu çalışmada 12 haftalık 230-280 gr ağırlığındaki 20 dişi sıçan 5 gruba ayrılmıştır: kontrol, sham, SK (20 mg/kg, intraperitoneal), GB (80 mg/kg/gün, oral) ve SK+GB (SK, 20 mg/kg; GB, 80 mg/kg/gün). Çiftleşmeden sonra hem SK hem de SK+GB grupları tek doz SK'ya maruz bırakıldı. Doğumdan sonra her gruptan rastgele 8 dişi sıçan seçildi ve 32 gün boyunca normal koşullarda standart yem ile beslendi. Deneme süresinin sonunda, sıçanlara anestezi altında kardiak perfüzyon uygulandı. Sonrasında beyincikleri çıkarıldı ve mikroskop altında beyincikten elde edilen kesitler sterolojik ve histopatolojik olarak incelendi.
Stereolojik bulgular, SK grubunda toplam Purkinje hücre sayısının kontrol grubuna göre anlamlı derecede azaldığını göstermiştir (p<0,05). Ayrıca SK grubunda ortalama moleküler tabaka, granüler tabaka, ak madde ve beyincik hacimleri kontrol grubuna göre anlamlı olarak azalmıştır (p<0,05). Buna ek oarak, SK+GB grubunda toplam Purkinje hücre sayısı, ortalama moleküler tabaka, granüler tabaka, ak madde ve beyincik hacimleri SK grubuna göre anlamlı derecede artmıştır (p<0,05). Histopatolojik değerlendirme de beyincik dokularının yapılarında değişiklikler göstermiştir.
SK'ya prenatal maruziyet, sıçan beyinciklerinde yapısal değişikliğe neden olmuştur. Ayrıca GB uygulanmasının, SK'nın beyincik üzerindeki olumsuz etkilerini anlamlı olarak azalttığı gözlemlendi.
ABSTRACT
This study aimed to investigate the negative effects of prenatal exposure to cyclophosphamide (SK) on the rat cerebellum. In addition, we evaluated the protective activity of ginkgo biloba (GB) against possible biological changes caused by SK in cerebellar tissues. In the present study, 20 adult female rats (12 weeks old, weighing 230-280 g) were divided into 5 groups: control, sham, SK (20 mg/kg, intraperitoneal), GB (80 mg/kg/day, orally), and SK+GB (SK, 20 mg/kg; GB, 80 mg/kg/day). After mating, both SK and SK+GB groups were exposed to SK. After parturition, 8 female rats were randomly selected from each group, maintained under normal conditions, and fed with standard chow for 32 days. At the end of the experimental period, rats underwent cardiac perfusion under anesthesia. The cerebella were immediately removed, and sections obtained from the cerebellum were examined at the light microscope stereologically and histopathologically.
Stereological findings showed that the total number of Purkinje cells in the SK group was significantly decreased compared to the control group (p<0.05). In addition, the mean volumes of molecular layer, granular layer, white matter, and cerebellum were significantly decreased in the SK group compared to the control group (p<0.05). In addition, total Purkinje cell number, mean molecular layer volume, granular layer volume, white matter volume and cerebellum volume were significantly increased in the SK+GB group compared to the SK group (p<0.05). Histopathological evaluation also exhibited changes in the structures of the cerebellum tissues.
Our results showed that prenatal exposure to SK caused structural changes in rat cerebellum. It was also observed that GB significantly reduced the negative effects of SK on the cerebellum. |
