Abstract:
ÖZET
Bu tez kapsamında, dopamin tayini için karbon nanofiber (CNF), magnetit nanopartiküller (Fe3O4) ve gümüş nanopartiküller (AgNP) ile modifiye edilmiş camsı karbon elektrot (GCE) temelli bir elektrokimyasal sensör geliştirildi. Tez çalışmasında, GCE yüzeyini modifiye etmede kullanılan malzemelerin miktarları optimize edilerek elektrodun optimum yüzey bileşimi belirlendi. AgNP/CNF-Fe3O4/GCE’nin hazırlanması sırasında, her bir modifikasyon basamağında elde edilen modifiye elektrotların elektrokimyasal karakterizasyonu dönüşümlü voltametri (CV) yöntemi ile, modifiye elektrotların yüzey karakterizasyonu ise taramalı elektron mikroskopisi (SEM) ile incelendi. Elektrokimyasal sensör için çalışma pH’sı ve çalışma potansiyeli optimize edildi ve optimum pH 8,5; optimum potansiyel ise +0,25 V olarak belirlendi. AgNP/CNF-Fe3O4/GCE’nin performans özellikleri optimum koşullarda incelendi ve dopamine 0,20 μM ile 546,5 μM arasında 37,24 μA mM-1 duyarlılıkla cevap verdiği belirlendi. AgNP/CNF-Fe3O4/GCE’nin tekrar kullanılabilirlik, tekrar üretilebilirlik, kararlılık, seçicilik gibi diğer performans özellikleri de incelendi. AgNP/CNF-Fe3O4/GCE’nin gerçek numunelerde dopamin tayininde kullanılabilirliğini belirlemek amacıyla, dopamin hidroklorür içeren iki farklı enjeksiyon numunesinde dopamin tayini yapıldı. Geri kazanım değerleri %99,8 ile %102,0 arasında bulundu.
ABSTRACT
In this thesis, an electrochemical sensor based on glassy carbon electrode (GCE) modified with carbon nanofiber (CNF), magnetite nanoparticles (Fe3O4) and silver nanoparticles (AgNP) was developed for dopamine determination. In the thesis study, the optimum surface composition was determined by optimizing the amount of material used to modify the GCE surface. During the preparation of AgNP/CNF-Fe3O4/GCE, the electrochemical characterization of the modified electrodes obtained at each modification step was performed by cyclic voltammetry (CV) method, and the surface characterization of the modified electrodes was performed by scanning electron microscope (SEM). The working pH and working potential were optimized fort he electrochemical sensor and the optimum pH was determined as 8.5; the optimum potential was determined as +0.25 V. The performance characteristics of AgNP/CNF-Fe3O4/GCE were examined under optimum conditions and it was determined that it responded to dopamine between 0.20 μM and 546.5 μM with a sensitivity of 37.24 μA mM-1. Other performance properties of AgNP/CNF-Fe3O4/GCE such as reusability, reproducibility, stability, selectivity were also investigated. In order to determine the usability of AgNP/CNF-Fe3O4/GCE in the determination of dopamine in real samples, dopamine was determined in two different injection samples containing dopamine hydrochloride. Recovery values were determined between 99.8% and 102.0%.
