Öğe Acrylamide applied during pregnancy causes the neurotoxic effect by lowering BDNF levels in the fetal brain(Pergamon-Elsevier Science Ltd, 2018) Erdemli, Mehmet Erman; Aladag, M. Arif; Altinoz, Eyup; Demirtas, Sezin; Turkoz, Yusuf; Yigitcan, Birgul; Bag, Harika GozukaraObjectives: The aim of this study is to elucidate the possible mechanism of neurotoxic effect of acrylamide (AA) applied during pregnancy on fetal brain development and to show the effect of N-acetylcysteine (NAC) on AA toxicity. Materials and methods: Four groups were formed with 9 pregnant rats each as control (C), acrylamide (AA), N-acetylcysteine (NAC), acrylamide plus N-acetylcysteine (AA plus NAC) groups. Caesarian section was implemented on the 20th day of pregnancy. Malondialdehyde (MDA), reduced glutathione (GSH), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), catalase (CAT) and Brain-derived neurotrophic factor (BDNF) levels were analyzed and histopathologic examinations were performed in brain tissues of the fetuses. Results: Our data indicated that AA caused necrotic death and hemorrhagic damages in fetal brain tissue with decreasing BNDF levels and increasing oxidative stress. N-acetylcysteine prevented the toxic effects of its on fetal brain (p < 0.05). Conclusion: Our study indicated that acrylamide has toxic effects in the fetal brain and N-acetylcysteine prevents its toxic effect.Öğe Ameliorative effects of crocin on tartrazine dye-induced pancreatic adverse effects: a biochemical and histological study(Springer Heidelberg, 2021) Erdemli, Zeynep; Altinoz, Eyup; Erdemli, Mehmet Erman; Gul, Mehmet; Gozukara, Harika; Gul, SemirThe present study aimed to analyze the impact of tartrazine (T) and crocin (Cr) applications on the pancreas tissues of the Wistar rats. A total of 40 Wistar rats were randomly divided into 4 groups with 10 rats in each group, including the Control, T, Cr, and T + Cr groups. After 3 weeks of application, the pancreatic tissues of the rats were removed under anesthesia and rat blood samples were obtained. Tissues were analyzed with biochemical and histopathological methods. It was determined that T administration increased malondialdehyde (MDA), total oxidant status (TOS), oxidative stress index (OSI), glucose, triglyceride, LDL, VLDL, and total cholesterol levels. However, it decreased reduced glutathione (GSH), total antioxidant status (TAS), superoxide dismutase (SOD), catalase (CAT), and HDL levels when compared with the other groups. It was observed that Cr administration significantly increased GSH, SOD, CAT, TAS, and HDL levels when compared with the control group. In the T group, histopathological changes were observed in pancreatic tissue, leading to damages in exocrine pancreas and islets of Langerhans and increased caspase-3 immunoreactivity (p <= 0.001). Co-administration of Cr and T brought the biochemical and histopathological findings closer to the control group levels. The administration of T induced damage in the pancreas with the administered dose and frequency. Cr can increase the antioxidant capacity in pancreas tissue. Co-administration of T and Cr contributed to the reduction of the toxic effects induced by T. Materials and Methods: Ethics committee approval was obtained and 30 Wistar rats were randomly divided into 3 groups of 10 rats: Control (C), Periodontitis (P), and Periodontitis + Crocin (P + Cr). After the treatments, rat kidney tissues were incised under anesthesia and blood samples were collected. Biochemical and histopathological analyses were conducted on the samples. Results: Malondialdehyde (MDA), total oxidant status (TOS), and oxidative stress index (OSI) increased in P group rat kidney tissues; urea, creatinine, Tumor necrosis factor-alpha (TNF-alpha), interleukin-6 (IL-6), and interleukin 1 beta (IL-1 beta) levels increased in the serum; glutathione (GSH), superoxide dismutase (SOD), catalase (CAT) and total antioxidant status (TAS) levels were reduced in rat kidney tissues, and renal histopathology deteriorated. In the P + Cr group, we observed improvements in biochemical and histopathological parameters when compared with the P group. Conclusion: Periodontitis (P) led to deterioration in oxidative stress parameters and histopathology by increasing the oxidants in kidney tissue. P also led to inflammation in the blood of the rats. Periodontitis + Crocin (P + Cr) administration alleviated the effects of P due to powerful antioxidant anti-inflammatory properties. Cr could be employed as a protective agent in P-induced inflammation and oxidative damage.Öğe Antioxidant, anti-inflammatory, and anti-apoptotic effects of crocin against doxorubicin-induced myocardial toxicity in rats(Springer Heidelberg, 2021) Abdulkareem Aljumaily, Sara Asaad; Demir, Mehmet; Elbe, Hulya; Yigitturk, Gurkan; Bicer, Yasemin; Altinoz, EyupDoxorubicin (DOX) is a well-known chemotherapeutic drug for most malignancies including breast cancer and leukemia whilst the usage of DOX is limited owing to its cardiotoxicity. In the present study, we aimed to investigate the effects of crocin on doxorubicin-induced cardiotoxicity in rats. Forty rats were randomly divided into four groups: (a) control [received normal saline as a dose of 1 ml/kg by intraperitoneal injection (ip) for 15 days], (b) crocin (received crocin as a dose of 40 mg/kg/24h by ip for 15 days), (c) DOX (received DOX as a dose of 2 mg/kg/48h by ip in six injection, cumulative dose 12 mg/kg), and (d) DOX+crocin (received DOX as a dose of 2 mg/kg/48h by ip in six injection, and crocin as a dose of 40 mg/kg/24h i.p for 15 days). As compared to the controls, the results showed that DOX administration caused significant increases in lipid indices [triglyseride (TG), low-dencity lipoproteins (LDL) (p<0.001), and very low-dencity lipoproteins (VLDL) (p<0.005)], oxidative stress parameters [malondialdehyde (MDA) and total oxidant status (TOS) (p<0.001)] and cardiac markers [creatine kinase-muscle/brain (CK-MB) and cardiac troponin I (cTnI) (p<0.001)]. Besides, significant decreases in antioxidant defense systems [glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), and total antioxidant status (TAS) (p<0.001)] were observed. The present study also demonstrated that co-administration of crocin with DOX significantly ameliorated the lipid profile (p<0.005), cardiac markers (p<0.005), and oxidative stress indices (p<0.001) as compared to DOX group. Histopathologically, significant increase in the mean histopathological damage score (MHDS) was found in the DOX group as compared to the controls (p<0.001). In contrast, the administration of crocin with DOX alleviated MHDS in myocardium (p<0.001). Taken together, our results reveal that crocin might be a cardioprotective agent in DOX-treated patients for cancer.Öğe Biochemical investigation of the toxic effects of acrylamide administration during pregnancy on the liver of mother and fetus and the protective role of vitamin E(Taylor & Francis Ltd, 2017) Erdemli, Mehmet Erman; Altinoz, Eyup; Aksungur, Zeynep; Turkoz, Yusuf; Dogan, Zumrut; Bag, Harika GozukaraObjectives: To investigate the toxic effects occurring in the liver tissues of the pregnant rats and the fetuses, which are administered acrylamide and vitamin E as a protector during pregnancy. Materials and methods: This research was conducted with the permission of Laboratory Animals Ethical Board of Inonu University Faculty of Medicine. Forty rats, of which their pregnancy is validated via vaginal smear, were distributed into five different groups. On the 20th day of pregnancy, pregnant rats and fetuses are decapitated. Malondialdehyde (MDA), reduced glutathione (GSH), total antioxidant status (TAS), total oxidant status (TOS) and xanthine oxidase (XO) levels were measured in the liver samples taken from mother and fetuses. Results: It was detected that acrylamide administered during pregnancy increased MDA, TOS, XO levels statistically significantly and decreased the GSH level (p <= 0.05) in the pregnant rat liver tissue when compared to all other groups. In the vitamin E administered group; GSH, TAS levels significantly increased statistically and TOS and XO levels dropped to levels of the control group (p <= 0.05), in comparison to all other groups. Among all groups, no biochemical changes were observed in the fetus liver tissue (p > 0.05). Conclusion: The liver of pregnant rats functions as a protective pre-filter by detoxifying acrylamide effectively and the acrylamide that reaches fetus liver is detoxified by the cytochrome P-450 system of the fetus liver. To be able to figure out the biochemical mechanism, more advanced studies are needed.Öğe Can crocin play a preventive role in Wistar rats with carbon tetrachloride-induced nephrotoxicity?(Mashhad Univ Med Sciences, 2018) Erdemli, Mehmet Erman; Gul, Mehmet; Altinoz, Eyup; Aksungur, Zeynep; Gul, Semir; Bag, Harika GozukaraObjective(s): To investigate protective role of crocin by attempting to create nephrotoxicity with carbon tetrachloride. Materials and Methods: Ethics committee approval was obtained and 50 male Wistar rats were randomly divided into 5 groups that included 10 rats each: Control, Corn oil, Crocin, Carbon tetrachloride (CCl4), and Crocin + Carbon tetrachloride. Following the experiments, the rats were decapitated under anesthesia and incised kidney tissues were subjected to biochemical and histological examinations. Results: In the CCl4 administered group, MDA, TOS, Bun, and creatinine levels increased, GSH, SOD, CAT, and TAS levels decreased (P <= 0.05), glomerular collapse in kidney sections, narrowing and local occlusion in Bowman's space in certain glomeruli, inflammatory cell infiltration and congestion were observed when compared to all other groups. There was a significant decrease in increased MDA, TOS, Bun, and creatinine levels, and a significant increase in decreased GSH, SOD, CAT, and TAS levels in CCl4 + crocin administered group compared to the CCl4 group (P <= 0.05), local minimal glomerular damage, tubular damage, inflammatory infiltration, and vascular collagen symptoms were observed in kidney sections, however significant improvement was observed in damage findings when compared to the CCl4 group. Conclusion: At this dose and time interval, against a highly toxic chemical such as CCl4, crocin was able to suppress oxidative stress by playing a protective role in the kidney tissue.Öğe Crocin (active constituent of saffron) improves CCl4-induced liver damage by modulating oxidative stress in rats(Walter De Gruyter Gmbh, 2019) Cosgun, Binnaz Elif; Erdemli, Mehmet Erman; Gul, Mehmet; Gul, Semir; Bag, Harika Gozukara; Erdemli, Zeynep; Altinoz, EyupBackground: CCl4 is a widely used xenobiotic for the purpose of causing liver damage in experimental studies. In this study, we aimed to reveal the effects of crocin on liver injury caused by CCl4 via free radical scavenging properties. Materials and methods: Animals were divided into five groups of 10: control; corn oil; crocin; CCl4 ; CCl4 + crocin. Tissue samples were carefully removed and separated for biochemical and histological investigations. Results: CCl4 administration led to significant increases in MDA, SOD, CAT and TOS in liver tissue, and AST, ALT and ALP levels in plasma (p <0.05). In addition, CCl4 caused significant decreases in GSH and TAS (p < 0.05). When animals were treated with crocin, high MDA, SOD, CAT, TOS levels, and AST and ALP activities decreased and GSH and TAS levels increased. Control group exhibited normal histological appearance; however extensive necrosis areas were detected in the CCl4 group. In the CCl4 +crocin group, pathological changes were markedly decreased and the appearance of liver tissue was almost similar to the control groups. Conclusion: Our results showed that crocin suppresses oxidative stress with antioxidant properties and has a protective effect on tissue damage caused by CCl4.Öğe Crocin attenuates oxidative and inflammatory stress-related periodontitis in cardiac tissues in rats(Wroclaw Medical Univ, 2021) Kocaman, Gulhan; Altinoz, Eyup; Erdemli, Mehmet Erman; Gul, Mehmet; Erdemli, Zeynep; Zayman, Emrah; Bag, Harika Gozde GozukaraBackground. Periodontitis, a chronic inflammatory disease affecting the supporting tissues around the teeth, causes significant inflammatory and oxidative changes in cardiac tissue. Crocin is the active constituent of Crocus sativus (saffron) which has antioxidant properties and is protective against cardiovascular disturbances. Objectives. The present study aimed to investigate the protective effects of crocin on periodontitis-induced oxidative/inflammatory cardiac degeneration in rats in vivo. Materials and methods. Thirty female Sprague Dawley rats were randomly divided into 3 groups: control group, periodontitis group (PD) and periodonditis plus crocin group (PD+Cr). Experimental periodontitis was induced by placing silk ligatures on the maxillary second molar teeth for 30 days. Afterward, crocin (100 mg/kg body weight/day) was administered to the PD+Cr group and saline was administered to the PD group and the control group for 15 days. The subjects were sacrificed on the 45th day. Results. Histological and biochemical analyses demonstrated that inducing periodontitis caused obvious damage to cardiac tissues which was significantly ameliorated by crocin (p < 0.05). Significant improvements in bone resorption parameters (cross-linked C-telopeptide of type I collagen and bone alkaline phosphatase) were also observed in the PD+Cr group (p < 0.05). In addition, crocin caused significant reductions of malondialdehyde levels and total oxidant score while antioxidant levels (glutathione, superoxide dismutase, total antioxidant score, and catalase) were significantly higher in PD+Cr group (p < 0.05). Conclusions. This study reveals that periodontitis may cause oxidative damage in cardiac tissue and crocin improves periodontitis-induced degenerative changes in heart tissue, which is associated with its antioxidant properties.Öğe Crocin protects intestine tissue against carbon tetrachloride-mediated oxidative stress in rats(Aepress Sro, 2018) Cosgun, Binnaz E.; Erdemli, Mehmet E.; Gul, Mehmet; Gul, Semir; Bag, Harika G.; Aksungur, Zeynep; Altinoz, EyupSaffron is used in traditional medicine for its hypolipidemic, anti-inflammatory and anti carcinogenic properties as a natural remedy in treatment of diseases. The objective of the present study was to demonstrate the protective effect of crocin (one of the main ingredients of saffron) on carbon tetrachloride (CCl4) damage in intestinal mucosa. MDA, GSH, SOD, CAT, TAS and TOS levels were measured in experimental animal tissue samples and these were compared with histologic lesions induced by CCl4.CCl4 caused an increase in MDA, SOD, CAT and TOS levels and a significant decrease in GSH and TAS levels in rat intestinal tracts. After crocin treatment, serious improvements were observed in histological lesions and biochemical results in the intestinal tract. In conclusion, crocin inhibited the toxic effects induced by CCl4 in the intestine by its strong antioxidant properties.Öğe Crocin treatment exerts anti-inflammatory and anti-oxidative effects in liver tissue damage of pinealectomized diabetic rats(Springer Heidelberg, 2023) Karayakali, Melike; Altinoz, Eyup; Elbe, Hulya; Koca, Oguzhan; Onal, Melike Ozgul; Bicer, Yasemin; Demir, MehmetDiabetes mellitus (DM) is a chronic metabolic disorder with an increasing global prevalence that leads to significant morbidity and mortality. The liver plays a vital role in glycemic regulation in physiological and pathological conditions such as DM. Free radical formation and inhibition of antioxidant defense systems play a role in the liver damage pathogenesis in diabetic patients The antioxidant, anti-diabetic, anti-inflammatory, and radical scavenging properties of crocin are known. This study was designed to determine the possible protective effects of crocin against liver tissue damage in pinealectomized diabetic rats. Sixty rats were divided into six groups: Control, Sham+streptozotocin (STZ), Pinealectomy (PINX), PINX+STZ, PINX+Crocin, and PINX+STZ+Crocin. PNX procedure was carried out on the first day of the experiment. Intraperitoneal (i.p.) injection of 50 mg/kg STZ was performed on the 30th day of the experiment to induce DM. Crocin (50 mg/kg; i.p.) was applied for 15 days after the pinealectomy procedure and induction of DM. Crocin decreased the markers (alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), interleukin-1 beta (IL-1 beta), and malondialdehyde (MDA)) of liver damage and increased antioxidant enzyme levels and tissue total antioxidant status. Histological results showed that the administration of crocin exhibited a protective effect against liver damage caused by STZ. These results indicate that crocin evidence protection against liver injury caused by STZ.Öğe Crocin, the compound of the dried stigma of Crocus sativus L (saffron), restores doxorubicin-induced disturbances in kidney functioning, oxidative stress, inflammation, renal tissue morphology and TGF-? signalling pathways(Taylor & Francis Ltd, 2024) Altinoz, Eyup; Cetinavci, Dilan; Abdulkareem Aljumaily, Sara Asaad; Elbe, Hulya; Cengil, Osman; Bicer, YaseminDoxorubicin (Dox), an anthracycline antibiotic, is a chemotherapeutic drug for several cancer treatments. However, its clinical usage has been restricted because of severe side effects, including nephrotoxicity. This study aimed to demonstrate the possible nephroprotective effects of crocin (Cr) against Dox-induced oxidative stress, renal inflammation, renal morphology and transforming growth factor-beta (TGF-beta) signalling pathways in Dox-exposed rats. Hence, the rats were injected for 15 d consecutively with saline, six different injections of Dox until the cumulative dose reached 12 mg/kg., daily Cr (40 mg/kg), and Dox + Cr combination. Cr increased the activities of superoxide dismutase (SOD) and catalase (CAT), GSH content and suppressed inflammation and oxidative stress in Dox-exposed rats. Our results were confirmed by immunohistochemical findings that Cr treatment ameliorates the expressions of IL1 beta and TGF-beta in Dox-induced nephrotoxicity. Conclusionally, Cr exhibits adequate nephroprotective effects against Dox-induced nephrotoxicity on rat kidney architecture and tissue function by stabilising cellular redox homeostasis, reducing renal fibrosis and suppressing inflammation. [GRAPHICS] .Öğe Cytotoxic effect of extract from Dunaliella salina against SH-SY5Y neuroblastoma cells(General Physiol And Biophysics, 2015) Ataseyer-Arslan, Belkis; Yilancioglu, Kaan; Bekaroglu, Maide G.; Taskin, Emre; Altinoz, Eyup; Cetiner, SelimCytotoxic effects of essential oils extracted from Dunaliella sauna on SH-SY5Y human neuroblastoma cells were investigated in this study. GC-MS analysis was used for determination of the composition of essential oils found in Dunaliella salina extract. All experimented concentrations of Dunaliella salina extract on SH-SY5Y human neuroblastoma cells were significantly more cytotoxic than the tested concentrations of the extract on ECV304 human endothelial cells used as a control. Fifthy compounds were detected in GC-MS analysis of the extract, and five major compounds were predominantly found as follows: octadecanoic acid, methyl ester (27.43%); hexadecanoic acid, methyl ester (Cas) methyl palmitate (24.82%); 9,12,15-octadecatrienoic acid, ethyl ester, (Z,Z,Z)- (7.39%); octadecanoic acid (5.03%), pentadecanoic acid (3.60%). The cytotoxic activity of Dunaliella salina extract on SH-SY5Y human neuroblastoma cells might be due to high concentrations of octadecanoic acid and hexadecanoic acid. Furthermore, results indicate that the extract demonstrates some proliferative effect on ECV304 cells in a dose-dependent manner between 0.25 and 5 mu g/ml. These results suggest that Dunaliella sauna may have anticancer potential against human neuroblastoma cells.Öğe Effect of crocin on oxidative stress in recovery from single bout of swimming exercise in rats(General Physiol And Biophysics, 2016) Altinoz, Eyup; Ozmen, Tarik; Oner, Zulal; Elbe, Hulya; Erdemli, Mehmet E.; Bag, Harika G.Physical exercise could cause muscle and tissue damage due to increase in the formation of free oxygen radicals during exercise. The aim of the present study was to investigate the effect of crocin on parameters associated with oxidative stress in recovery from acute swimming exercise in rats. Rats were divided into eight groups; Normal Control (NC, untreated and did not swim), Crocin Control (CC, received crocin and did not swim), Exe-1, Exe-24, Exe-48 (untreated and swam) and and Exe-Cro-1, Exe-Cro-24, Exe-Cro-48 (received crocin and swam). AST, ALP, LDH, CK, XO enzymes levels increased after swimming in untreated and crocin-treated groups, but there was a less increase in crocin-treated groups. The highest MDA levels in serum were determined in Exe-1 compared with all other groups. There was significant difference between control and exercise groups in MDA level (p = 0.033). In contrast, there was significant difference between control and exercise groups in GSH level (p < 0.001). In addition, crocin given to swimming rats significantly increased GSH levels (p < 0.05) and decreased MDA levels when compared with untreated exercise groups. In conclusion, crocin is able to protect liver and skeletal muscle tissue against exercise-induced oxidative damage by preventing reactive oxygen species (ROS) production.Öğe THE EFFECT OF SAFFRON (ITS ACTIVE CONSTITUENT, CROCIN) ON THE CARDIOVASCULAR COMPLICATION AND DYSLIPIDEMIA IN STREPTOZOTOCIN INDUCED DIABETIC RATS(African Networks Ethnomedicines, 2015) Altinoz, Eyup; Taskin, Emre; Oner, Zulal; Elbe, Hulya; Arslan, Belkis AtaseverBackground: Diabetes mellitus (DM) causes serious complications such as coronary heart disease, atherosclerosis, nephropathy, retinopathy and neuropathy. Materials and Methods: Rats were randomly divided into three groups, each containing 10 rats: control group, DM group; DM+crocin group. Normal saline was administered in the control and DM groups, and crocin was administered in DM+crocin group at a dose of 20 mg/kg bw/day for 21 days. Trunk blood and the hearth tissue were collected for histopathological and biochemical examination. Results: DM led to increment of MDA levels (p<0.05) and the serum levels of total cholesterol, TG, LDL and VLDL (p<0.05) and decrease of GSH levels (p<0.01) compared to control group in serum and heart tissue. Diabetic rats were treated with crocin, MDA levels (p<0.05) and the levels of total cholesterol, TG and VLDL decreased significantly, but GSH levels (p<0.05) increased remarkably compared to DM rats. Statistically significant increase in histopathological damage score was found in the DM group (p<0.0001). Histopathological changes markedly regressed in DM+crocin group. Conclusion: Our results showed that crocin might prevent diabetes induced cardiovascular complications by reduction of oxidative stress and dyslipidemia.Öğe The effects of acrylamide and vitamin E on kidneys in pregnancy: an experimental study(Taylor & Francis Ltd, 2019) Erdemli, Mehmet Erman; Aksungur, Zeynep; Gul, Mehmet; Yigitcan, Birgul; Bag, Harika Gozukara; Altinoz, Eyup; Turkoz, YusufObjectives: The objective of this study is to investigate possible damages to kidney tissues of pregnant rats and their fetuses exposed to acrylamide during pregnancy and possible protective effects of vitamin E against these damages. Material and methods: Rats were randomly assigned to five groups of control, corn oil, vitamin E, acrylamide, vitamin E + acrylamide, six pregnant rats in each. Mother and fetal kidney tissues were examined for malondialdehyde (MDA), reductase glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), total antioxidant status (TAS), total oxidant status (TOS), urea, creatine, trace elements such as Zn and Cu in the serum and histopathological analyses were conducted. Results: It was determined that acrylamide, administered during pregnancy, statistically significantly increased MDA and TOS levels, maternal serum urea, creatinine, and Zn levels, while it decreased GSH, TAS, SOD, and CAT levels (p <= .05) when compared with all other groups in the kidney tissues of pregnant rats and their fetuses and caused tubular degeneration, hemorrhage, narrowing, and closure in Bowman's space, and, in the E vitamin group, it statistically significantly increased GSH, TAS, SOD, CAT, urea, creatinine, and Zn levels when compared with other groups and lowered TOS and MDA levels to those of the control group (p < .05) and there were no differences between the groups histologically. Methods: Thirty animals randomized in three groups containing ten animals in each group as follows; control (non-diabetic rats), DM (STZ-induced untreated diabetic rats), DM+crocin (STZ-induced diabetic rats treated with crocin,). Crocin was given at a dose of 20 mg/kg bw/day by gavage for 21 days. Results: STZ injection caused a significant increase in mRNA expression of antioxidant enzymes, SOD, CAT and GPX when compared to control group. Crocin given to diabetic rats significantly decreased mRNA expression of antioxidant enzymes, SOD, CAT and GPX when compared to DM group. Conclusion: The present study demonstrates that crocin can modulate mRNA expression of antioxidant enzymes, SOD, CAT and GPX and oxidative stress in the brain of the STZ induced diabetic rats.Öğe Effects of pinealectomy and crocin treatment on rats with isoproterenol-induced myocardial infarction(Taylor & Francis Ltd, 2022) Demir, Mehmet; Altinoz, Eyup; Elbe, Hulya; Bicer, Yasemin; Yigitturk, Gurkan; Karayakali, Melike; Ballur, Arwa Fadil HaqiThe present study aimed to analyze the effects of pinealectomy and crocin treatment in isoproterenol-induced myocardial damage. Seventy rats were divided into seven groups: control, sham control, pinealectomy (PNX), isoproterenol (ISO; 85 mg/kg on the 29th and 30th days of the experiment, subcutaneous injection), PNX + ISO, PNX + crocin (50 mg/kg/day for 30 days, intragastric administration), and PNX + ISO + crocin. PNX procedure was performed on the first day of the study. A significant increase was observed in serum cardiac damage markers (CK-MB, Troponin I) after ISO administration. ISO administration led to a significant increase in cardiac oxidative stress parameters, such as malondialdehyde (MDA) and total oxidant status (TOS), while it led to a decrease in antioxidant defense system parameters, such as reduced glutathione (GSH), superoxide dismutase (SOD), catalase (CAT) and total antioxidant status (TAS) when compared to control groups. Elevated MDA and TOS levels were observed, while reduced SOD and CAT activities, and decreased GSH and TAS levels were observed in the group that underwent PNX and ISO administration when compared to the PNX group. Furthermore, in the PNX + ISO + Crocin group, SOD and CAT activities, and GSH and TAS levels ameliorated and MDA and TOS levels were reduced with the crocin treatment when compared to the PNX + ISO group. Also, marked increases were observed in serum cardiac markers, histopathological and immunohistochemical findings after the crocin treatment. All findings demonstrated that crocin could be employed as a cardioprotective agent due to its antioxidant, anti-inflammatory, and anti-apoptotic properties.Öğe The effects of pinealectomy and melatonin treatment in acrylamide-induced nephrotoxicity in rats: Antioxidant and anti-inflammatory mechanisms(Pergamon-Elsevier Science Ltd, 2024) Demir, Mehmet; Altinoz, Eyup; Cetinavci, Dilan; Elbe, Hulya; Bicer, YaseminObjective: Acrylamide (AA) is toxic and forms in food that undergoes high-temperature processing. This study aimed to investigate the effects of AA-induced toxicity on renal tissue in pinealectomized rats and the possible protective effect of exogenous Melatonin (ML) administration. Materials and Methods: Sixty rats were randomized into 6 groups (n = 10): Sham, Sham+AA, Sham+AA+ML, PX, PX+AA, and PX+AA+ML. Sham and pinealectomized rats received AA (25 mg/kg/day orally) and ML (0.5 ml volume at 10 mg/kg/day, intraperitoneal) for 21 days. Results: The results showed that malondialdehyde (MDA), total oxidant status (TOS), oxidative stress index (OSI), tumor necrosis factor-alpha (TNF-alpha), and interleukin 113 (IL-113) levels of the kidney and urea and creatinine levels of serum in the PX (pinealectomy)+AA group were more increased than in the Sham+AA group. In addition, glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), and total antioxidant status (TAS) levels decreased more in the PX+AA group than in the Sham+AA group. Also, we observed more histopathologic damage in the PX+AA group. On the other hand, up-regulation of kidney tissue antioxidants, down-regulation of tissue oxidants, and improvement in kidney function were achieved with ML treatment. Also, histopathological findings such as inflammatory cell infiltration, shrinkage of glomeruli, and dilatation of tubules caused by AA toxicity improved with ML treatment. Conclusion: ML supplementation exhibited adequate nephroprotective effects against the nephrotoxicity of AA on pinealectomized rat kidney tissue function by balancing the oxidant/antioxidant status and suppressing the release of proinflammatory cytokines.Öğe Hepatoprotective effects of crocin on biochemical and histopathological alterations following acrylamide-induced liver injury in Wistar rats(Elsevier France-Editions Scientifiques Medicales Elsevier, 2017) Gedik, Sema; Erdemli, Mehmet Erman; Gul, Mehmet; Yigitcan, Birgul; Bag, Harika Gozukara; Aksungur, Zeynep; Altinoz, EyupThe objective of the present study is the treatment of oxidative damage caused by acrylamide induced oxidative stress in rats with the administration of a strong antioxidant, namely crocin. High acrylamide (AA) levels have genotoxic, carcinogenic and neurotoxic effects on living organisms. In the present study, 40 Wistar rats were randomly divided into four equal groups. These groups were control, acrylamide (25 mg/kg), crocin (50 mg/kg), acrylamide + crocin (25 mg/kg acrylamide and 50 mg/kg crocin) groups. At the end of the application, biochemical and histological variations were examined in liver and blood samples. It was observed that acrylamide administration significantly decreased liver GSH and TAS levels when compared to the control group. On the contrary, it was also observed that AST, ALT, ALP, SOD and CAT activities and TOS and MDA levels increased as a result of acrylamide administration. Histopathological examinations demonstrated inflammatory cell infiltration, hepatocellular necrosis and hemorrhage areas in AA group liver sections. Furthermore, intracytoplasmic vacuolization was detected in hepatocytes. After crocin treatment, it was observed that GSH and TAS levels increased while AST, ALT, ALP, SOD and CAT activities and TOS and MDA levels decreased. Significant decreases were observed in inflammatory cell infiltration and vascular congestion in liver sections and intracytoplasmic vacuolization in hepatocytes after the crocin treatment, while no hepatocellular necrosis and hemorrhages were observed. In the present study, it was demonstrated that crocin treatment removed acrylamide induced liver damage due to the strong antioxidant properties of crocin.Öğe Influence of Pinealectomy and Long-term Melatonin Administration on Inflammation and Oxidative Stress in Experimental Gouty Arthritis(Springer/Plenum Publishers, 2022) Ballur, Arwa Fadil Haqi; Altinoz, Eyup; Yigitturk, Gurkan; Onal, Melike Ozgul; Elbe, Hulya; Bicer, Yasemin; Karayakali, MelikeGout is an inflammatory arthritis characterized by the deposition of monosodium urate (MSU) crystals in the joints or soft tissue. MSU crystals are potent inflammation inducers. Melatonin (MLT) is a powerful endogenous anti-inflammatory agent and effective in reducing cellular damage. In the present study, possible underlying mechanisms associated with anti-inflammatory and antioxidative effects were investigated in rats with gouty arthritis and melatonin deprivation treated with MLT. Fifty-six rats were divided into seven groups: control, sham control, pinealectomy (PNX), MSU (on the 30th day, single-dose 20 mg/ml, intraperitoneal), MSU + MLT (10 mg/kg/day for 30 days, intraperitoneal), MSU + PINX and MSU + PINX + MLT. PNX procedure was performed on the first day of the study. As compared to the controls, the results showed that MSU administration caused significant increases in oxidative stress parameters (malondialdehyde and total oxidant status). Besides, significant decreases in antioxidant defense systems (glutathione, superoxide dismutase and total antioxidant status) were observed. A statistically significant increase was found in the mean histopathological damage score in the groups that received MSU injection. It was found that histopathological changes were significantly reduced in the MSU + MLT group given MLT. In our study, it was determined that many histopathological changes, as well as swelling and temperature increase in the joint, which are markers of inflammation, were significantly reduced with MLT supplementation. These results suggest that melatonin ameliorates MSU-induced gout in the rat through inhibition of oxidative stress and proinflammatory cytokine production.
