Öğe The morphometrical and immunohistochemical investigation of the effect of topiramate on liver and the role of neuropeptide Y receptor in an obese female rat(Comenius Univ, 2020) Yahyazadeh, A.; Altunkaynak, B. Z.; Alkan, IOBJECTIVE: We aimed to investigate the possible effect of topiramate (TOP, 0.02 mg/kg/day) on the livers in a high-fat diet (HFD)-induced obesity rat model. The other objective was to evaluate the relationship between TOP administration and NPY level using anti-NPY1R antibody. METHODS: Twenty-four adult female Wistar albino rats were randomly assigned into four equal groups as follow: control (CONT), obese (OBS), TOP, and OBS+TOP. All liver samples were investigated using the stereological analysis, as well as immunohistochemical and histopathological examination. RESULTS: The total number of hepatocytes was significantly decreased in the OBS+TOP group compared to the CONT group or the OBS group (p < 0.05). We found a significant increase in the mean volume of liver in the OBS group compared to the CONT group (p < 0.05). Also, the mean volume of liver was significantly decreased in the OBS+TOP group compared to the OBS group (p < 0.05). CONCLUSION: Taken together, our findings suggest that decreased liver volume is possibly attributed to TOP administration via setting the NPY level in the obese rats. Further, the side effects of TOP in combination with health risk of obesity may have led to an increase in hepatotoxicity and the subsequent hepatocyte loss (Fig. 7, Ref. 56). Abdominal adhesions are common and often develop after abdominal surgery. Currently, there are no effective targeted pharmacotherapies for treating adhesive disease. In this regard, ginger is wildly used in traditional medicine because of its anti-inflammatory and antioxidant effects and has been investigated for peritoneal adhesion treatment. This study analyzed ginger ethanolic extraction via HPLC to have a 6-gingerol concentration. Four groups induced peritoneal adhesion to evaluate ginger's effects on peritoneal adhesion. Then, ginger extract (50, 150, and 450 mg/kg) was administered by gavage in various groups of male Wistar rats (220 ± 20 g, 6-8 weeks). After scarifying the animals for biological assessment, macroscopic and microscopic parameters were determined via scoring systems and immunoassays in the peritoneal lavage fluid. Next, the adhesion scores and interleukin IL-6, IL-10, tumor necrosis factor-(TNF-) ?, transforming growth factor-(TGF-) ?1, vascular endothelial growth factor (VEGF), and malondialdehyde (MDA) were elevated in the control group. The results showed that ginger extract (450 mg/kg) notably decreased inflammatory (IL-6 and TNF-?), fibrosis (TGF-?1), anti-inflammatory cytokine (IL-10), angiogenesis (VEGF), and oxidative (MDA) factors, while increased antioxidant factor glutathione (GSH), compared to the control group. These findings suggest that a hydro-alcoholic extract of ginger is a potentially novel therapeutic strategy for inhibiting adhesion formation. Also, it might be considered a beneficial anti-inflammatory or antifibrosis herbal medicine in clinical trials. However, further clinical studies are required to approve the effectiveness of ginger. © 2023 Roghayeh Yahyazadeh et al.Öğe Protective effect of melatonin on the rat lung following exposure to 900-MHz electromagnetic field: a stereological and histopathological study(Ondokuz Mayis Universitesi, 2021) Yahyazadeh, A.; Kivrak, E.G.; Koç, G.E.; Altunkaynak, B.Z.Long-term use of cell phone emitting electromagnetic field (EMF) has increased concerns regarding histopathological change in vital organs. Our present study was aimed to investigate the effect of 900 megahertz (MHz) EMF on the rat lungs, as well as the possible efficacy of melatonin (MEL) on lung tissues. Fifteen female Wistar albino rats were randomly selected and assigned into five groups as follow: control (CONT), SHAM, EMF, melatonin (MEL) and EMF+MEL group. Subsequently, female rats were then mated, and pregnant rats underwent the experimental application for 21 days. After parturition, 6 pups (2 pups from each mother) were randomly chosen and maintained for 4 weeks. At the end of the 4th week, the lung tissues of all groups were used for the stereological analysis and histopathological examination. We found that the mean volumes of the alveoli, bronchioles and blood vessels were significantly lower in the EMF group compared to the control group (p<0.05). In the EMF+MEL group, the mean volumes of the alveoli, bronchioles and blood vessels were significantly increased compared to the EMF group. Our histopathological results showed marked change in the lung tissues. We speculated that exposure to EMF caused the damage to the rat lung tissues, and that MEL administration alleviated 900 MHz EMF-induced complications. © 2021 Ondokuz Mayis Universitesi. Z.Increasing cell phone use calls for clarification of the consequences of long term exposure to electromagnetic fields (EMF). We investigated the effects of EMF on the testes of 12-week-old rats as well as possible protective effects of luteolin on testis tissue. Twenty-four Wistar albino rats were randomly divided into four groups: control, EMF, luteolin, and EMF + luteolin. The number of Leydig cells, primary spermatocytes and spermatids were reduced in the EMF group compared to the control group. In the EMF + luteolin group, the number of Leydig cells, primary spermatocytes and spermatids was significantly greater than the EMF group. We found an increase in superoxide dismutase (SOD) activity in the EMF group compared to the control group. In the EMF group, we found decreased wet weight of testes and serum testosterone levels compared to the control group. Decreased SOD enzyme activity, and increased serum testosterone levels and weight of the testes were observed in the EMF + luteolin group compared to the EMF group. EMF also affected sperm morphology. We found that in rat testis repeated exposure to 900 MHz EMF caused changes in testicular tissue and that the antioxidant, luteolin, substantially reduced the deleterious effects of EMF.Öğe Stereological and histological assessment of the umbilical cord in new-born rat(Wolters Kluwer Medknow Publications, 2021) Altunkaynak, B.Z.; Yahyazadeh, A.Background: Umbilical cord plays a crucial role in the continuation of pregnancy by transferring nutrition and oxygen across the placenta to the fetus. We aimed to investigate the morphometrical and histological features of the umbilical cords in new-born rats. Materials and Methods: The adult male and female rats were chosen for matting purpose in the present study. Briefly, ten adult Wistar albino rats (female, n = 5; male, n = 5) were randomly assigned into five groups of two animals (female, n = 1; male, n = 1). Immediately after parturition, two umbilical cords of new-born rats (0-day-old) from each group were randomly selected. Finally, ten umbilical cord samples were examined using the histological and stereological methods in the light and electron microscopes. Results: The total numbers of Hofbauer cells and mesenchymal stromal cells was estimated statistically. We also calculated the mean volume of umbilical cords, arteries and veins, as well as arterial and venous lumens. Our histological findings also exhibited the histological features of Hofbauer cells, mesenchymal stromal cell cells, and blood vessels. Conclusion: Our findings showed more detailed information about umbilical cord tissues and their components, and that may contribute to the diagnose of umbilical cord complications in the developing fetus. © 2020 Journal of Microscopy and Ultrastructure
